Mechanism of Action
Palbociclib is an oral, selective cyclin-dependent kinase (CDK) 4 and 6 inhibitor. It blocks CDK4/6-mediated phosphorylation of the retinoblastoma (Rb) protein, preventing progression from the G1 to S phase of the cell cycle. This results in cell-cycle arrest and inhibition of proliferation in Rb-positive cancer cells, particularly hormone receptor–positive breast cancer.
