Cyclosporin is a calcineurin inhibitor that suppresses T-cell mediated immune response.
It binds to cyclophilin, forming a complex that inhibits calcineurin phosphatase activity.
This inhibition blocks IL-2 transcription, reducing T-lymphocyte activation and proliferation.
It is primarily used in organ transplantation and autoimmune diseases.
Indications include prevention of graft rejection and treatment of severe psoriasis, rheumatoid arthritis, and nephrotic syndrome.
